Results

High production yield for antibody

The results presented indicate that HYPE-293™ Transfection Kit (HYPE-293 + B293) achieves high antibody production level and largely outperforms other commercially available transfection reagents in 293-F cells growing in suspension with chemically defined medium. After 6 days in culture more than 600 mg/L of antibody can be produced under standard bioproduction process (30mL scale).

 

Fig.1: IgG Rabbit production after transient transfection of 293-F cells with HYPE-293 kit in 30 mL cell culture volume. Transfections were performed using 1.5 µg of plasmid DNA (antibody-expressing positive control vector, ThermoFisher) per mL of cell culture. 2 µL of HYPE-293 reagent per µg of plasmid was used. 293-F cells were cultured in 30 mL of medium in 125 mL shaken flasks. B293 (1X final) was added to the cells just after the addition of the complexes (10µL in 1 mL). Density of cell culture was 1x106 cells/mL the day of transfection. Day 3 and day 6 post-transfection, 500µL of supernatant was collected and IgG was measured with Easy-Titer Rabbit IgG Assay Kit (ThermoFisher).

 

Outperforms Competitors

The results presented below indicate that HYPE-293™ Transfection Kit (HYPE-293 + B293) achieves the highest protein yield in comparison to other commercially available transfection reagents in 293-F cells growing in suspension with chemically defined medium.

SEAP was produced by transient transfection using HYPE-293 Transfection Kit, Free Style Max transfection reagent, LP2K, FHD, 293Fectin or IT-PRO transfection reagents according to manufacturer’s instructions. 293-F cells were cultured in 1 mL volume in a 24-well shaken plate Density of cell culture was 1x106 cells/mL the day of transfection. Day 3 post-transfection, 20µL of supernatants were collected and SEAP activity was measured using SEAP assay Kit (#SP00500).

 

Easy and efficient scale-up

The results indicate that protein production in 293-F suspension cells can be easily scaled up by using HYPE-293™ Transfection Kit. It has been used in volume up to 300 mL of cell culture without any protocol modification and protein yield production is even higher when compared to lower cell culture volume.

Transfections were performed using 0.75 or 1.5 µg of plasmid DNA per mL of cell culture. Ratio of 2 µL of HYPE-293 reagent per µg of plasmid was used. Density of cell culture was 1x106 cells/mL the day of transfection.
