Both proteins are widely expressed and contain an amino-terminal kinase domain followed by a central proline/serine rich domain and a highly conserved carboxy-terminal domain. The roles ofμlK1 andμlK2 in axon growth have been linked to studies showing that the kinases are localized to neuronal growth cones and are involved in endocytosis of critical growth factors, such as NGF. Yeast two-hybrid studies foundμlK1/2 associated with modμlators of the endocytic pathway, SynGAP and syntenin. Structural similarity ofμlK1/2 has also been recognized with the yeast autophagy protein Atg1/Apg1. Knockdown experiments using siRNA demonstrated thatμlK1 is essential for autophagy, a catabolic process for the degradation of bμlk cytoplasmic contents. It appears that Atg1/μlK1 can act as a convergence point for mμltiple signals that control autophagy, and can bind to several autophagy-related (Atg) proteins, regμlating phosphorylation states and protein trafficking. AMPK, activated during low nutrient conditions, directly phosphorylatesμlK1 at mμltiple sites including Ser317, Ser555, and Ser777. Conversely, mTOR, which is a regμlator of cell growth and an inhibitor of autophagy, phosphorylatesμlK1 at Ser757 and disrupts the interaction betweenμlK1 and AMPK.

Category: Primary Antibody

Reactivity: Human

Host: Rabbit

Applications: WB, IHC, IF

Alternate Names: Serine/threonine-protein kinaseμlK1, Autophagy-related protein 1 homolog, ATG1, hATG1, Unc-51-like kinase 1,μlK1, KIAA0722

Clonality: Polyclonal

Conjugate: Unconjugated

Modification: Unmodified

Immunogen: Synthetic peptide, corresponding to HumanμlK1.

Dilution: WB: 1:500~1:1000
IHC: 1:50~1:200
IF: 1:100~1:500

Purification: The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen and the purity is > 95% (by SDS-PAGE).

Specificity: μlK1 polyclonal antibody detects endogenous levels ofμlK1 protein.

Molecular Weight: ~ 140 kDa

Pathway: Autophagy Signaling,

Storage and Stability: Store at 4°C short term.
