Furthermore, hnRNP U has been shown to bind chromatin to help regulate its structure during interphase. During prophase, hnRNP U becomes phosphorylated by Aurora-B, which evicts hnRNP U and the chromatin-associated RNAs bound to it, a process critical to proper chromosome alignment. hnRNP U plays a role in the chromatin localization of Xist RNA, and loss of hnRNP U in ES cells fail to achieve an inactivated X chromosome. hnRNP U has also been shown to play a key role in immunity, functioning as a viral dsRNA sensor. Upon viral recognition, hnRNP U oligomerizes and activates numerous antiviral genes, including IFNB1 .
