RPA is a single-stranded DNA binding protein, whose DNA binding activity is believed to reside entirely in the 70 kDa subunit. The complex is required for almost all aspects of cellular DNA metabolism such as DNA replication, recombination, cell cycle and DNA damage checkpoints, and all major types of DNA repair including nucleotide excision, base excision, mismatch, and double-strand break repairs. In response to genotoxic stress in eukaryotic cells, RPA has been shown to associate with the Rad9/Rad1/Hus1 checkpoint complex. RPA is hyperphosphorylated upon DNA damage or replication stress by checkpoint kinases including ataxia telangiectasia mutated (ATM), ATM and Rad3-related (ATR), and DNA-dependent protein kinase (DNA-PK). Phosphorylation of RPA32 occurs at serines 4, 8, and 33. Hyperphosphorylation may alter RPA-DNA and RPA-protein interactions. In addition to the checkpoint partners, RPA interacts with a wide variety of protein partners, including proteins required for normal replication such as RCF, PCNA, and Pol α, and also proteins involved in SV40 replication, such as DNA polymerase I and SV40 large T antigen.

Purification and Purity: The antibody was purified by immunogen affinity chromatography.

Storage and Stability: Store at 4°C short term. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.

Alternative Name: REPA1; RPA70; Replication protein A 70 kDa DNA-binding subunit; RP-A p70; Replication factor A protein 1; RF-A protein 1; Single-stranded DNA-binding protein

Immunogen: Purified recombinant human RPA70 protein fragments expressed in E.coli.

Conjugate: Unconjugated

Modification: Unmodified

//cdn.shopify.com/s/files/1/0305/9482/6376/products/thumbnail_d1447bac803ec9be_11-14-09-19-51_9dd6b674-9fa5-4638-9f30-915e9d18e291.jpg?v=1706095083
