Background: Anoctamin 1, also called DOG1 for discovered on gastrointestinal stromal tumors protein 1 or TMEM16A for Transmembrane Member 16A, is a plasma membrane calcium-activated chloride channel. It is essential for chloride secretion from epithelial tissues, and has been shown to be a specific marker for cells of Cajal, the pacemaker cells that control smooth muscle contraction. Anoctamin 1 knockout mice exhibit an altered gastric smooth muscle rhythmic contraction. More recently, research studies have identified Anoctamin 1 as a heat sensor in nociceptive neurons. Heat above 44ºC triggers anoctamin 1-dependent depolarization, contributing to the mediation of thermal nociception.

Extra Notes: Western blot analysis of ANO1 expression in LO2 (A), HeLa (B), mouse lung (C), rat skeletal muscle (D) whole cell lysates., Immunofluorescent analysis of ANO1 staining in U2OS cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a Alexa Fluor 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.
