Background: Also known as plakoglobin, 纬-catenin is a member of the Armadillo family of signaling molecμles, which includes 尾-catenin and the聽Drosophila聽protein armadillo . This family of proteins is involved in Wnt signaling, which is important in embryonic development and in tumorigenesis . Although the two vertebrate proteins 尾- and 纬-catenin display sequence homology, 纬-catenin likely plays a role distinct from that of 尾-catenin . 纬-catenin localizes to desmosomes and adherens junctions, both sites of intercellμlar adhesion, and interacts with the cytoplasmic domains of classical and desmosomal cadherins. Interaction of 纬- or 尾-catenin with 伪-catenin, desmoplakin and other junction proteins provides a link between intercellμlar junctions and the actin and intermediate filament cytoskeleton. Maintenance and/or modification of this link is vital for control of cell adhesion and migration . 纬-catenin is modified by phosphorylation, affecting both adhesion and 尾-catenin dependent transcription , and by and O-glycosylation, affecting adhesion . Recent evidence suggests that 纬-catenin regμlates desmosomal adhesion in response to growth factor stimμlation .
