Background: The mTORC1 kinase complex is a critical regulator of cell growth. Its activity is modulated by growth factors and nutrients including amino acids. MAP4K3 is a mediator between nutrient signal and mTORC1. Research studies suggest that amino acid sufficiency leads to the phosphorylation of Ser170 on MAP4K3, which activates mTORC1 .

Extra Notes: Western blot analysis of GLK expression in MCF7 (A) whole cell lysates., Immunohistochemical analysis of GLK staining in human skeletal muscle formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
