Background: Interferon-induced 15 kDa protein (ISG15) acts as ubiquitin by conjugating to intracellular target proteins such as JAK1 or MAPK3/ERK1 through an enzyme pathway distinct from that of ubiquitin. ISG15 shows specific chemotactic activity towards neutrophils and activates them to induce the release of eosinophil chemotactic factors. ISG15 is also involved in paracrine, autocrine and endocrine mechanisms by inducing IFN-g secretion by monocytes and macrophages, as in cell-to-cell signaling. ISG15 is a cytoplasmic protein expressed mainly in muscle, epithelial, neuronal and lymphoid cells.

Purification and Purity: This antibody is purified through a protein G column.

Storage and Stability: Store at 4°C short term. Aliquot and store at -20°C long term. Avoid freeze-thaw cycles.

Specificity: Recognizes endogenous levels of ISG15 protein.

Extra Notes: Western blot analysis of ISG15 expression in Hela (A), MCF7 (B) whole cell lysates., Immunofluorescent analysis of ISG15 staining in Hela cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
