Background: The protein encoded by this gene is a member of the dual specificity protein kinase family, which acts as a mitogen-activated protein (MAP) kinase kinase. MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals. This protein kinase lies upstream of MAP kinases and stimulates the enzymatic activity of MAP kinases upon wide variety of extra- and intracellular signals. As an essential component of MAP kinase signal transduction pathway, this kinase is involved in many cellular processes such as proliferation, differentiation, transcription regulation and development.

Purification and Purity: The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen and the purity is > 95% (by SDS-PAGE).

Storage and Stability: Store at 4°C short term. Aliquot and store at -20°C long term. Avoid freeze-thaw cycles.

Extra Notes: Western blot analysis of extracts of various cell lines, using Phospho-MEK1-S298 antibody at 1:1000 dilution or MEK1 antibody. HeLa cells were treated by PMA/TPA at 37℃ for 15 minutes after serum-starvation overnight or treated by ATP at 30℃ for 1 hour.
Secondary antibody: HRP Goat Anti-Rabbit IgG at 1:10000 dilution.
Lysates/proteins: 25ug per Lane.
Blocking buffer: 3% BSA.
Detection: ECL Basic Kit.
Exposure time: 1s., Western blot analysis of extracts of various cell lines, using Phospho-MEK1-S298 antibody at 1:1000 dilution. C2C12 cells were treated by CIP at 37℃ for 1 hour or treated by ATP at 30℃ for 1 hour. C6 cells were treated by CIP at 37℃ for 1 hour or treated by ATP at 30℃ for 1 hour.

Western blot was performed from the immunoprecipitate using Phospho-MEK1-S298 pAb at a dilution of 1:1000. 293T cells were treated by PMA/TPA at 37℃ for 30 minutes after serum-starvation overnight.
