Background: The prohibitins, called PHB1 and PHB2, are highly conserved proteins that are present in multiple compartments in eukaryotic cells. PHB1 is 30kDa tumor suppressor protein involved in cell cycle control. PHB1 has been found in mitochondria, the nucleus and the plasma membrane, as well as extracellularly in circulation. In mitochondria prohibitins mainly exist as membrane-bound ring complexes and function as chaperones maintaining mitochondrial protein stability during protein synthesis and transportation. In the nucleus prohibitins interact with transcription factors such as Rb and p53 to regulate target gene transcription. Extracellular prohibitin can bind and activate C3 to enhance complement activation.

Extra Notes: Western blot analysis of Prohibitin expression in Hela (A), A549 (B), HL60 (C), U2OS (D), C6 (E) whole cell lysates., Immunohistochemical analysis of Prohibitin staining in human cholangiocarcinoma formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.48). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX., Immunofluorescent analysis of Prohibitin staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.
