SREBP-1c Polyclonal Antibody Product: Liquid in 0.42% Potassium phosphate, 0.87% Sodium chloride, pH 7.3, 30% glycerol, and 0.01% sodium azide. Catalogue Numbers: BS67746-50, BS67746-100 Sizes: 50µl, 100µl Swiss-Prot: P36956 Host: Rabbit Reactivity: Human, Mouse, Rat Applications: WB, IHC Background: The low density lipoprotein (LDL) receptor mediates the endocytic uptake of cholesterol-carrying lipoproteins, thereby controlling cholesterol levels in cells and plasma. Transcription of the LDL receptor gene is controlled by a ten base pair sequence in the 5' flanking region, designated sterol regulatory element 1 (SRE-1). When cellular sterol stores are depleted, the element is activated, the gene is transcribed and the cellular uptake of LDL increases. A set of SREbinding proteins (SREBPs) have been identified, including two basic helixloop-helix leuicine zipper (bHLH-Zip) transcription factors, designated SREBP-1 and SREBP-2. SREBP-1 (also designated ADD1, for adipocyte determination and differentiation factor) is synthesized as a precursor that is attached to the nuclear envelope and endoplasmic reticulum. In sterol-depleted cells, the membrane-bound precursor is cleaved to generate a soluble NH2-terminal fragment that translocates to the nucleus to activate transcription. Sterols inhibit the cleavage of SREBP-1. Product: Liquid in 0.42% Potassium phosphate, 0.87% Sodium chloride, pH 7.3, 30% glycerol, and 0.01% sodium azide. Purification and Purity: The antibody was purified by immunogen affinity chromatography. Specificity: Recognizes endogenous levels of SREBP-1c protein. Note: For research use only, not for use in diagnostic procedure. Extra Notes: Western blot analysis of SREBP-1c expression in mouse kidney (A), rat brain (B) whole cell lysates., Immunohistochemical analysis of SREBP-1c staining in rat liver formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. Bioworld Molecular Weight: ~ 72 kDa Storage and Stability: Store at 4°C short term. Aliquot and store at -20°C long term. Avoid freeze-thaw cycles. Alternative Name: BHLHD1; SREBP1; Sterol regulatory element-binding protein 1; SREBP-1; Class D basic helix-loop-helix protein 1; bHLHd1; Sterol regulatory element-binding transcription factor 1 Immunogen: Recombinant protein corresponding to human SREBP-1c.
