Background: Survivin is a 16 kDa anti-apoptotic protein highly expressed during fetal development and cancer cell malignancy . Survivin binds and inhibits caspase-3, controlling the checkpoint in the G2/M-phase of the cell cycle by inhibiting apoptosis and promoting cell division . This regulatory process requires the phosphorylation of survivin at Thr34 by p34 cdc2 kinase . Gene targeting using a Thr34 phosphorylation-defective survivin mutant, as well as antisense survivin, have been shown to inhibit tumor growth .

Purification and Purity: The antibody was purified by immunogen affinity chromatography.

Specificity: Recognizes endogenous levels of Survivin protein.

Extra Notes: Western blot analysis of Survivin expression in Jurkat (A) whole cell lysates., Immunofluorescent analysis of Survivin staining in MCF7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.
