Background: Proliferating cell nuclear antigen (PCNA) is a member of the DNA sliding clamp family of proteins that assist in DNA replication. Crystal structure data suggests that a PCNA homotrimer ring can encircle and slide along the DNA double helix. Multiple proteins involved in DNA replication, DNA repair, and cell cycle control bind to PCNA rather than directly associating with DNA, thus facilitating fast processing of DNA. PCNA protein expression is a well-accepted marker of proliferation.
Purification and Purity: The antibody was purified by immunogen affinity chromatography.

Specificity: Recognizes endogenous levels of PCNA protein.

Extra Notes: Western blot analysis of PCNA expression in Hela (A), NIH3T3 (B), COS7 (C), C6 (D), CHOK1 (E) whole cell lysates., Immunofluorescent analysis of PCNA staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.
