Lena Biosciences offers a variety of in vitro services using their innovative SeedEZ and PerfusionPal products
The assays are run in the proprietary SeedEZ scaffold that enables both cadherine and integrine signaling in 3D. Common assays include: CellTiter-Glo, alamarBlue, LDH release, etc. Assays are routinely run with primary cells and iPSCs and Lena Biosciences has extensive experience with brain co-cultures, liver, cancers such as breast, liver, lung etc.
Lena Biosciences generates advanced 3D tissue models using co-cultures of primary cells or cell lines based on your specifications, or using cells provided by you. Models may be perfused (interstitial perfusion) and vascularized in their proprietary PerfusionPal system to model, for example, vascularized tumors.
Neuronal assays are run in 2D and 3D. Testing may also be performed in their cortical model comprising tri-culture of neurons, astrocytes and microglia. Cells may be further supported by the Blood Substitute that increases cellular respiratory metabolism several-fold and reduces cell death. Cultures can also be perfused in the proprietary PerfusionPal organ-on-a-chip-like system providing month-long support.
Phase I drug metabolism services are performed using perfused 3D cultures of primary human hepatocytes with log-fold increase in CYP450 activity. CYP450 assays are run using primary human hepatocytes in 2D and 3D.
Short-term and long-term DILI using primary liver cells with log-fold increase in Phase I drug metabolism and multifold increase in cell respiratory metabolism.
Toxicity assays are offered in 2D and 3D, and in advanced, synthetically vascularized models supported by the Blood Substitute and interstitial perfusion. Assays may be performed using a client's cells and standard assays (CellTiter-Glo or total LDH, kinetic LDH release etc.) For immune cell therapy toxicity, immune cells may be co-cultured with the organ models, or the organ models exposed to activated immune cell medium to assess toxicity in vitro.
Off-target toxicity is evaluated in the 12-well organ-on-a-chip-like system PerfusionPal. For in vitro cell therapy testing the toxicity of activated immune cells is measured after co-culture with 3D human organ models, or after incubation of 3D organ models in activated immune cell medium.
