Background: In eukaryotes, the phosphorylation and dephosphorylation of proteins on serine and threonine residues is an essential means of regulating a broad range of cellular functions, including division, homeostasis and apoptosis. A group of proteins that are intimately involved in this process are the protein phosphatases. In general, the protein phosphatase (PP) holoenzyme is a trimeric complex composed of a regulatory subunit, a variable subunit and a catalytic subunit. Four major families of protein phosphatase catalytic subunit have been identified, designated PP1, PP2A, PP2B (calcineurin) and PP2C.An additional protein phosphatase catalytic subunit, PPX (also known as PP4) is a putative member of a novel PP family. The PP1 family is comprised ofsubfamily members PP1α, PP1β and PP1γ, which are MgATP-dependent enzymes. PP1 inactivity is maintained through its association with the inhibitory protein NIPP-1 (nuclear inhibitor of PP1). Phosphorylation of NIPP-1 by cAMP-PK or casein kinase II results in the release of active PP1.

Extra Notes: Western blot analysis of PP1 beta expression in K562 (A), rat brain (B), C6 (C), NIH3T3 (D), Hela (E) whole cell lysates., Immunohistochemical analysis of PP1 beta staining in human colon cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.61). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX., Immunofluorescent analysis of PP1 beta staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a AF488-conjugated secondary antibody (green) in PBS at room temperature in the dark.
